When a suppository is exposed to body temperature it melts and the active in the suppository must either coat the local mucosal membranes (if it is delivering an active that works on the surface of the mucosa) or it must penetrate through the membranes into the blood stream (if it is delivering a drug for systemic activity). In either case, effective dispersal of the drug is important.
Emulsifiers can be used to effectively disperse lipophilic material and drugs in the primarily aqueous point of delivery, by forming micelles or stabilizing small droplets. BASF has a selection of compendial, GMP-manufactured emulsifiers. Polysorbate 60 (Kolliphor® PS 60) and Polysorbate 80 (Kolliphor® PS 80), both have a history of use in suppositories.
