Suppositories: Solubilizers and Solvents

BASF offers a variety of solubilizers and solvents ideal for suppository mass formulations.

Suppositories have a rigid structure so that they can be properly handled and inserted during application, but they still must have a degree of softness and lubricity, and must contain solubilized or dispersed API. These attributes are governed by the selection of and amount of fluid solubilizers and solvents that are used in the formulation.

In anhydrous, hydrophobic suppositories the solubilizer is usually an oil. Medium chain triglycerides (Kollisolv® MCT 70)octyldodecanol (Kollicream® OD)isopropyl myristate (Kollicream® IPM), and other oils, can all be used to provide lubricity, increase softness and aid in the solubilization of lipophilic drugs. Depending upon the physico-chemical properties of the API, these oils may also be able to accelerate the penetration of the drug through the mucosal membranes.

In hydrophilic suppository masses the same is accomplished by using propylene glycol (Kollisolv® PG) or other excipients like glycerine.
